Electron Microscopic Observation on the Cytochemical Localization and Phospholip se Activity in Candida species Isolated from Oral Cavity


Abstract
The phospholipase activity of 38 isolated of oral Candida species were determined by a plate assay and titrimetric method.
Ninety two percent of Candida albicans were phospholipase producers whereas none of the other Candida species produced the enzyme.
The degree of phospholipase activity (Pz value and IU/L) of individual isolates was constant despite the large variation in activity among different isolates.
Phospholipase production in vitro Vv as limited to a,narrow pH range (pH3. 0-4.0) and is suppressed by increasing pH in the edia.
The lethal dose 50 of phospholipas Candida albicans (Pz value = 0.39) in mice and chick embryo were 1.0x104 cells /ml, but inonproducing Candida albicans (Pz value= 1. 00) were 3. 7 X 105 cell /ml -4. 0 x 105 cell /ml in¢¥ mice and 1.0 x 106 cell /ml in chick embryo.
The electron microscopy of cytochemical localization of the Candida cellular phospholipase occurs in the cell membrane and walls of nondividing cells.
